Probiotics Supplements Reduce ER Stress and Gut Inflammation Associated with Gliadin Intake in a Mouse Model of Gluten Sensitivity

چکیده

Exposure to gluten, a protein present in wheat rye and barley, is the major inducer for human Celiac Disease (CD), chronic autoimmune enteropathy. CD occurs about 1% worldwide population, genetically predisposed individuals bearing leukocyte antigen (HLA) DQ2/DQ8. Gut epithelial cell stress innate immune activation are responsible breaking oral tolerance gliadin, gluten component. To date, only treatment available long-term gluten-free diet. Several studies have shown that an altered composition of intestinal microbiota (dysbiosis) could play key role pathogenesis through modulation permeability regulation system. Here, we show gliadin induces endoplasmic reticulum (ER) condition small intestine gluten-sensitive mouse model coadministration probiotics efficiently attenuates both unfolded response (UPR) gut inflammation. Moreover, formulations might differ their activity at molecular level, especially toward three axes UPR. Therefore, administration potentially represent new valuable strategy treat patients, such as those affected by CD.

BL-7010 Demonstrates Specific Binding to Gliadin and Reduces Gluten-Associated Pathology in a Chronic Mouse Model of Gliadin Sensitivity

Celiac disease (CD) is an autoimmune disorder in individuals that carry DQ2 or DQ8 MHC class II haplotypes, triggered by the ingestion of gluten. There is no current treatment other than a gluten-free diet (GFD). We have previously shown that the BL-7010 copolymer poly(hydroxyethyl methacrylate-co-styrene sulfonate) (P(HEMA-co-SS)) binds with higher efficiency to gliadin than to other proteins ...

Chemical chaperones reduce ER stress and adipose tissue inflammation in high fat diet-induced mouse model of obesity

Obesity, which is characteristic by chronic inflammation, is defined as abnormal or excessive fat accumulation in adipose tissues. Endoplasmic reticulum (ER) stress is increased in adipose tissue of obese state and is known to be strongly associated with chronic inflammation. The aim of this study was to investigate the effect of ER stress on adipokine secretion in obese mice and explore the po...

Assessment of Probiotics Mixture on Memory Function, Inflammation Markers, and Oxidative Stress in an Alzheimer\'s Disease Model of Rats

Background: The most important cause of neurodegeneration in Alzheimer's disease (AD) is associated with inflammation and oxidative stress. Probiotics are microorganisms that are believed to be beneficial to human and animals. Probiotics reduce oxidative stress and inflammation in some cases. Therefore, this study determined the effects of probiotics mixture on the biomarkers of oxidative stres...

Gluten sensitivity: from gut to brain.

Gluten sensitivity is a systemic autoimmune disease with diverse manifestations. This disorder is characterised by abnormal immunological responsiveness to ingested gluten in genetically susceptible individuals. Coeliac disease, or gluten-sensitive enteropathy, is only one aspect of a range of possible manifestations of gluten sensitivity. Although neurological manifestations in patients with e...

Effect of gluten free diet on immune response to gliadin in patients with non-celiac gluten sensitivity

BACKGROUND Non-celiac gluten sensitivity is a syndrome characterized by gastrointestinal and extra-intestinal symptoms occurring in a few hours/days after gluten and/or other wheat protein ingestion and rapidly improving after exclusion of potential dietary triggers. There are no established laboratory markers for non-celiac gluten sensitivity, although a high prevalence of first generation ant...
